Yes, one reading of 恋 is レン (ren) but here in the manga title it is 'koi' reading only. But anyway "hiren" is the correct current pronunciation, another form may be "himegoi" ひめごい while "hikoi" is not listed in dictionaries. The fact that the author choose to specify a furigana means she's using a personal reading which is not the common/current one(kind of the same thing they sometimes do with first names).
